A technique to prepare a thin film from soft materials (macro-molecule materials, biological materials, etc.), which are difficult to cut at room temperature. The specimen is cooled by liquid nitrogen to harden and is formed to a thin section using a cryo microtome. The achievable thickness of the section is several 10 nm to several μm.
It is noted that the exposed faces of the specimen can also be used for SEM observations.
