“Ultramicrotome” is a tool to prepare a thin film specimen for SEM / TEM.
For the blade, a glass knife or a diamond knife is used. A thin film specimen with a thickness of several 10 to several 100 nm is created by slicing with the knife, similarly to scraping with a plane. The ultramicrotome is suitable for slicing soft materials such as resin-embedded biological materials or macro-molecule materials. It can also be used to slice hard materials like metals. In the case of SEM, not only the sliced sections, but also exposed faces (square of several 100 μm to several mm) are subjected to observation.
