en bloc staining
En bloc staining is one of staining (electron staining) techniques for biological specimens. After postfixation, en bloc staining is carried out before dehydration. In the staining, a biological specimen (block tissue) is immersed in a solution of uranium acetate or lead aspartic acid. En bloc staining adheres heavy elements of uranium and/or lead to cell membranes and textures. Thus, absorption of the electrons to the heavy-element regions becomes large, resulting in dark-image formation. Consequently, the structures of the cell membranes and textures are clearly visualized in TEM images.
